Abstract
Description
The present invention relates to the development of a moisture stable and at the same time Enteric-coated oral dosage form for animals.
Fischimmunisierung praktisch aussichtslos [Dresenkamp, Dissertation Berlin 1992].
The manufacture of moisture-stable and at the same time gastric juice-resistant oral dosage forms is of great importance in the manufacture of veterinary medicinal products. An example of the need for such pharmaceutical forms is the combating of viral and bacterial diseases in fish in pond management, lake and river fishing. However, the development of vaccine formulations against viral diseases has so far been problematic. Due to the high expenditure and the short protective effect, passive fish immunization is practically hopeless [Dresenkamp, dissertation Berlin 1992]. Only active immunization with inactivated or live vaccines is considered promising. The following options are described in the literature.
Schäperclaus (fish diseases, Akademie Verlag, (1990)) and Smith (Vaccination against vibrosis in fish vaccination (Ellis, A. Ed., Academic press, London, 67-84, (1988)) report on immunization procedures using intrapertonic injections. The ones to be vaccinated Fish are immersed in a narcotic solution, on a table after the effects begin placed and vaccinated with dosing syringes. However, such an application method is in the intensive mass fish farming impractical and associated with very high costs. In addition comes the high stress associated with vaccination for the fish. Also be large amounts of antigen required. The reasons mentioned contribute to the fact that despite good The protective effect of vaccination for fish immunization was only very limited.
Technische Probleme (z. B. Flußhaltung) und Streßfaktoren begrenzen jedoch diesen Einsatz.2. The bath vaccination was successfully tested with inactivated bacterial vaccines (Kölbl, Tierärztliche Umschau 45, 624-629, 1990). To do this, put the vaccine in the fish tank given and the water supply and quantity throttled beforehand. Technical problems (e.g. River maintenance) and stress factors limit this use.
In a spray process that is also known, the fish are removed from the network and sprayed with an appropriate pressure (Gould et al. in Fish Pathol. 13, 63-68 (1978)). The method represents a high stress load for the fish. It also has the consequence that the Fish receive different amounts of vaccine and are therefore not always sufficient be immunized.
The oral application proves itself for mass vaccinations of animals of any size and especially suitable for young animals. The daily feeding schedule does not have to change so that the application is not associated with stress for the animals [Kolb, dissertation, Gießen, 1994]. Trying with gastric juice-insoluble coatings a drug form develop that allows immunization, however, was not sufficient because the gastric juice-resistant layer dissolved in the water after a short time and thus one effective immunization was prevented (Assmus and Schubert, dissertation Halle, 1980).
There are currently two options for administering the active ingredient or vaccine orally:
An attempt is made to add the active ingredient to the feed preparation. However, problems arise from the high Antigen content, which must be 100 times greater than with parenteral administration, since it is already in the In advance by mechanical barriers, release or destruction of the antigen in the water or in the Gastric effects are lost (Horsch, immunoprophylaxis in farm animals, Fischer Verlag, Jena, 1977).
The most important alternative currently is that of Stampniok (dissertation, Halle 1985) developed lipid pearl. An acceptable immunization was thus achieved to reach. The disadvantage of this pharmaceutical form results from the high cost of production.
The object of the present invention is therefore to provide novel and inexpensive substances develop that are superior to the above-mentioned prior art.
A substance will now be described which has the disadvantages of the already known substances overcomes.
The previous substances and processes have so far been inadequate due to high costs Possibility of transfer to large-scale production, high stress on the animals, limited uses in factory farming and inadequate protection of the Pharmaceutical form labeled in water.
The aim of the new substance is to immunize or give an oral application To achieve drug delivery. According to the invention this object is achieved in that it is the new substance for granules, pellets, powder or tablets for the treatment of animals acts as the outer layer at least one acid-labile or retarding coating have and in addition an acid-stable, but soluble at neutral or basic pH or contain swelling or retarding core or the core with an acid-stable, at neutral or basic pH value but soluble or swelling or retarding Material is covered. The outer layer preferably dissolves at a pH value of less than 5 or swells.
The basis of the new substance is a powder mixture, which is preferably made from potato starch and Lactose exists. However, they are also powder mixtures made from other substances such as starches or possible from feed bases. A solid active ingredient (e.g. Antibiotic or serum lyophilisate) added. Should this be unfavorable, then there is Possibility to add or dissolve the active ingredient or the serum in the binder. In addition, the serum can also be added directly to the powder and then by the Binders are solidified. They are used as binders in food chemistry and known pharmaceutical technology compounds such. B. PVP or gelatin Available.
With the aid of the binder, the powder mixture is thus solidified, whereby the particle size can be controlled by choosing the amount of liquid binder. Especially particles between 0.1 and 10 mm are favorable. The particle size can be changed that it comes closest to the food of the animals depending on their age and size. The Manufactured advantageously by granulators such. B. by a Fluid bed granulator or by a screen press granulator.
To realize the moisture and water stability as well as the acid stability According to the invention, various principles are available:
Abb. 1). Bei lösungsmittelempfindlichen Wirkstoffen oder Lebendimpfstoffen wird die
innere Hülle vorteilhafterweise als wäßrige Dispersion überzogen.
1. The outer shell is insoluble in the neutral and basic range, but swelling and acid-labile. The inner shell is acid-stable, but soluble in the neutral and basic area (see Fig. 1). In the case of active substances which are sensitive to solvents or live vaccines, the inner shell is advantageously coated as an aqueous dispersion.
The following materials are suitable for the outer shell:
Copolymers of acrylic acid (e.g. copolymers with a cationic character based on dimethylaminoethyl methacrylate and neutral methacrylic acid esters, copolymers with an anionic character based on methacrylic acid and methyl methacrylate, copolymers of acrylic acid and methyl methacrylate with a low content of ammonium groups).
Isopropanol, acetone and methylene chloride are preferred as solvents, or aqueous Dispersions used.
The following materials can be used for the inner shell:

b) Cellulosederivate (z. B. Hydroxyethylcellulose, Celluloseacetatphtalat,
Celluloseacetatsuccinat)
Als Lösungsmittel dienen Isopropanol, Aceton und Methylenchlorid.b) cellulose derivatives (e.g. hydroxyethyl cellulose, cellulose acetate phthalate, cellulose acetate succinate)
Isopropanol, acetone and methylene chloride are used as solvents. Diese Dispersionen liegen in wäßriger Form vor und können für lösungsmittelempfindliche Stoffe verwendet werden.c) methacrylic acid derivatives (e.g. polymers with anionic character based on Methacrylic acid and methyl methacrylate, anionic copolymers based on Methacrylic acid and ethyl acrylate, copolymers based on methacrylic acid and Ethyl acrylate, copolymers of methacrylic acid and methyl methacrylate and methyl acrylate). These dispersions are in aqueous form and can be used for solvent-sensitive Fabrics are used.

Als Lösungsmittel werden Isopropanol, Aceton und Methylenchlorid bzw. Gemische dieser bevorzugt.d) copolymers of acrylic acid
(e.g. copolymer with a cationic character based on dimethylaminoethyl methacrylate and neutral methacrylic acid esters, copolymer with an anionic character based on methacrylic acid and methacrylic acid methyl ester).
Isopropanol, acetone and methylene chloride or mixtures thereof are preferred as solvents.
The outer shell (see Fig. 2) is retarding in the neutral and basic area (delayed release) but swelling (see material under 1.). The inner shell is acid-stable, but soluble in the neutral and basic area (material see 2.). The following materials are used as the material for the outer shell:
-
a) Copolymerisate der Acrylsäure
(z. Als Lösungsmittel dienen Isopropanol, Aceton und Methylenchlorid.a) Copolymers of acrylic acid
(e.g. copolymers of acrylic acid and methacrylic acid methyl esters with a low content of ammonium groups). Isopropanol, acetone and methylene chloride are used as solvents.
The outer shell (see Fig. 3) is insoluble in the neutral and basic area, but swelling. Inside there is a matrix that ensures a delay or protection against the acid. Macromolecular substances such as starch, carboxymethyl starch, cellulose ether, alginates, galactomanan are used as the matrix material. There is also the option of incorporating acrylic acid derivatives (see 2.a and b) as scaffolding agents.
ESB, Varion Ad, Anionit AW17, Dowex1, Nalcite SBW, Duolite A42, Amberlite IR400 bzw.
CG50, Lewatit MN, Permutit E bzw. CCG, Lewatit MIH, Amberlite IR45, Duolite A6, A7.4. The outer shell is insoluble in the neutral and basic range, but swelling. Inside, in addition to the powder mixture and the active ingredient, there is a basic ion exchanger (resinate) that prevents premature release in an acidic environment ( Fig. 4).
The following substances are available as basic ion exchangers: Pemutit ESB, Varion Ad, Anionit AW17, Dowex1, Nalcite SBW, Duolite A42, Amberlite IR400 or CG50, Lewatit MN, Permutit E or CCG, Lewatit MIH, Amberlite IR45, Duolite A6 , A7.
It is possible to color the outer protective cover as desired. This makes it possible to adapt the color to the usual feed for the animals. Flavors can also be used additionally be placed on the protective cover so that the animal species is not between the dosage form and Can distinguish feeding.
The principles presented prove to be particularly advantageous for encapsulation gastric juice sensitive or acid labile drugs and vaccines. First of all, protection is guaranteed before moisture or water, there is no premature release such as for example with the pure feed-vaccine mixtures. It also prevents Live vaccines such as in the direct mixing of vaccines, in feed and thus can get directly into the environment. The production of the new oral dosage form is easily possible on an industrial scale. The fabrics are readily available. The application of the substances is also easily possible in factory farming.
B. Forellen), in Wasser flotierende Substanzen herzustellen. The new dosage form can be added to water like feed, but does not dissolve in it immediately. The composition can ensure that the substance can be fed according to the habits of the animals, for example, it is easily possible for animals that eat their food from the water surface (e.g. trout), in water to produce floating substances. Likewise, for animals that eat their food from the bottom, substances that sediment in water are easy to manufacture. The thickness of the water-insoluble The layer on the dosage form can be adapted to the necessary length of stay in the water. The Animals eat the particles that get into the stomach. The enteric coating, the matrix or the ion exchanger provides protection against the acid. The oral dosage form gets into the intestine. At a neutral pH, the layer, the active ingredients or, dissolves the vaccine will be released.
The present invention opens up various possibilities for administration a wide variety of medicines, including vaccines, high quality Feeds such as B. vitamins and minerals to a wide variety of animals. Especially it is suitable for use on fish.
The substances according to the invention are particularly suitable for animals as bait, for all types of feed admixtures or substances to be fed that are high in moisture are exposed or fed in a moistened or suspended state.
If the substance is exposed as bait in the forest, for example, it will rain or Moisture for no loss of active ingredient. The dosage form is eaten by the animal and after passing through the stomach without loss of active ingredient or titer (live vaccine) in the Gut where it releases the active ingredient at a neutral pH.
The following examples illustrate the invention without restricting it.
