Alle Probezähne wurden für die Dauer von 24 Stunden in einer RITC-Lösung (0,l%ige alkoholische Tetra- methylrhod-amin-Isothiocyanat-Lösung) gelagert. Anschlie- ßend wurde die Oberfläche mit Druckluft getrocknet (10 s) . Es folgte die Applikation eines mit RITC eingefärbten In- filtranten (Icon® Infiltrant, DMG) für 3 min und anschlie¬ ßende Lichthärtung für 60 s. Alle Proben wurden für 24 h in 30%igem H202 entfärbt, um RITC-Molkeküle (rot) die nicht im Infiltranten gebunden wurden zu entfernen. Es folgt eine Abschlusspolitur (4000 Körnung) . Zuletzt wurden alle Probenbereiche mit 0,1 % Natriumfluoresceinlösung (grün) 5 min lang gefärbt, überschüssige Lösung wurde unter fließendem Wasser entfernt. Die Analyse der Kunststoffpenetration er- folgte mit einem konfokalen Laser-Raster-Mikroskop (CLSM; Carl Zeiss) . Nicht mit Kunststoff infiltrierte Bereiche in Schmelzläsionen sowie das Dentin wurden aufgrund der Markierung mit Natriumfluorescein in grün dargestellt. Bereiche die der Infiltrant penetriert hat, wurden aufgrund des RITC-Farbstoffs rot dargestellt. Jede auszuwertende Fissur wurde standardisiert in drei Zonen (Fissurenspitze, Fissu¬ renbasis, Fissureneingang) eingeteilt. Die Abgrenzung von Fissurenspitze und Fissurenbasis wurde bei einer Fissuren¬ breite von 200 μιη angelegt, die von Fissurenbasis und Fis- sureneingang bei 1000 μιη. In jeder Zone wurden nun entlang des Übergangs vom Infiltranten zur Schmelzoberfläche 6 Messpunkte (links 3 und rechts 3) in gleichmäßigem Abstand voneinander bestimmt und Messlinien entlang des erkennbaren Schmelzprismenverlaufs angelegt. Somit wurden an insgesamt 18 gleichmäßig und standardisiert verteilten Messpunkten die Läsionstiefe und die Penetrationstiefe gemessen. Die absoluten Penetrationstiefen wurden in Relation zur jeweiligen Läsionstiefe gesetzt (prozentuale Penetration) und der statistischen Auswertung unterzogen (Kruskal-Wallis Test, Mann-Whitney Test; Signifikantniveau: p<0.05). The sample teeth were embedded in plastic blocks (Technovit, Heraeus Kulzer, Wehrheim) and test pieces were obtained with a thickness of 1,200 μιη each. Non-plastic infiltrated areas in enamel lesions as well as dentine were shown to be green due to labeling with sodium fluorescein. Areas that the infiltrant has penetrated were red due to the RITC dye. Each evaluated fissure was standardized into three zones (Fissurenspitze, Fissu ¬ renbasis, fissure) divided.
